چکیده

The proliferation and differentiation of normal granulocyte-macrophage (GM) progenitor cells is dependent on the presence of GM colony stimulating factor (GM-CSF). The proliferation of primary leukemic cells is also dependent on GMCSF, and some established myelomonocytic cell lines (e.g., WEHI3B D+) can be induced to differentiate by G-CSF. Using fluorescence-activated cell sorting, it has been possible to isolate highly purified granulocyte-macrophage colony forming cells (GM-CFC) [I I]. Fetal liver cells can be labeled with fluorescent pokeweed mitogen and the GM-CFC sorted on the basis of fluorescence intensity and light scatter. We have initiated an analysis of the 35S-labeled proteins in GM-CFC using two-dimensional gel electrophoresis [12]. In particular, the protein changes induced in GM-CFC by macrophage colony stimulating factor (M-CSF) [14], GM-CSF [4], and other hemopoietic regulators [5] have been monitored in an attempt to understand the extent and function of the protein synthetic events associated with commitment to differentiation or proliferation.

Effect of M20 interleukin-1 inhibitor on normal and leukemic human myeloid progenitors.

This study aimed to assess the effect of the M20 interleukin-1 (IL-1) inhibitor on normal and leukemic hematopoietic cells. The M20-derived IL-1 inhibitor was found to inhibit the growth of various hematopoietic cells. The in vitro proliferation of myeloid cell lines in serum-containing medium or proliferation of these cells induced by IL-1 in serum-free medium (measured by 3H-TdR) were inhibit...

In vivo control of differentiation of myeloid leukemic cells by recombinant granulocyte-macrophage colony-stimulating factor and interleukin 3.

The normal myeloid hematopoietic regulatory proteins include one class of proteins that induces viability and multiplication of normal myeloid precursor cells to form colonies (colony-stimulating factors [CSF] and interleukin 3 [IL-3], macrophage and granulocyte inducing proteins, type 7 [MGI-1]) and another class (called MGI-2) that induces differentiation of normal myeloid precursors without ...

Effects of human FLT3 ligand on myeloid leukemia cell growth: heterogeneity in response and synergy with other hematopoietic growth factors.

A novel hematopoietic growth factor for primitive hematopoietic progenitor cells, the ligand for the flt3/flk2 receptor, (FL), has been recently purified and its gene has been cloned. In the present study, we investigated the effects of FL on the proliferation and differentiation of normal and leukemic myeloid progenitor cells. We demonstrate that FL is a potent stimulator of the in vitro growt...

RAPID COMMUNICATION Granulocyte Colony-Stimulating Factor Activation of Stat3a and Stat3P in Immature Normal and Leukemic Human Myeloid Cells

Granulocyte colony-stimulating factor (G-CSF) is the cytokine critical for directing neutrophilic granulocyte differentiation. Acute myelogenous leukemia (AML) cells, which frequently arise from this lineage, respond aberrantly to G-CSF by proliferating without differentiating. The basis for this abnormal response is unknown. In the present study, we investigated whether G-CSF signaling in imma...

Selective regulation of the activity of different hematopoietic regulatory proteins by transforming growth factor beta 1 in normal and leukemic myeloid cells.

The viability of normal bone marrow myeloid precursor cells induced by interleukin-6 (IL-6) or IL-1 alpha and the ability of IL-6 and IL-1 alpha to induce the formation of colonies of granulocytes, macrophages, or megakaryocytes in densely seeded bone marrow cultures was suppressed by transforming growth factor-beta 1 (TGF-beta 1). Induction of normal bone marrow colony formation by IL-3 was mu...
